1. Factors Affecting Peptide Stability¶

2. Degradation Pathways¶
Hydrolysis¶
Peptide bond cleavage at Asp-X bonds. Accelerated by acidic pH and high temperature. Product: smaller peptide fragments. Prevention: Lyophilized storage, pH 4–6.
Oxidation¶
Reaction of Met, Cys, Trp, Tyr, and His with reactive oxygen species. Product: methionine sulfoxide, disulfide scrambling, crosslinking. Prevention: Antioxidants, nitrogen atmosphere, light protection.
Deamidation¶
Non-enzymatic hydrolysis of Asn and Gln side chains. Product: Asp, iso-Asp. Prevention: Low temperature, pH 4–6.
Aggregation¶
Non-covalent association of peptide molecules, often via hydrophobic interactions or β-sheet formation. Product: soluble and insoluble aggregates. Prevention: Avoid vortexing, low concentration, appropriate solvent.
3. Stability-Indicating Methods¶
RPL Peptide uses stability-indicating HPLC methods that can resolve the target peptide from all known and potential degradation products. System suitability requirements ensure method capability:
- Resolution from nearest degradation peak: ≥ 1.5
- Peak purity (diode array): ≥ 0.99
- Mass balance: 95–105%
